Cancer development and immune defense


"How does a dendritic cell, the professional antigen presenting cell in the immune system, exert it's actions, and how can we employ it to boost immune responses directed against tumors?". Martijn den Brok is searching for answers to this question using pre-clinical mouse studies, in which he provides dendritic cells with various stimuli derived from 'self' or 'non-self'. Of particular interest here are the stimuli that work together in synergy in the generation of in vivo immune responses. In his projects 'self' signals are given by injection of tumor-derived compounds, or by surgical techniques which destroy tumors in situ, like cryo-, radiofrequency-, radiation-, or HiFU-ablation. The 'foreign' signals are provided by administration of microbial TLR ligands or other adjuvants. Using various approaches the possible (molecular) working mechanisms of synergy are being connected to actual in vivo anti-tumor reactivity. The knowledge gathered in these studies is translated into clinical protocols using combination therapy with in situ tumor destruction in cancer patients.
